Here's an example of how to write an e-mail
Dividing into three parts: 20-40-40
1st paragraph should make up about 20% of your message, and the second and third paragraph
should make up about 40%.

This is an example of how your 1st paragraph should look like. Don't forget to write a brief introduction and your current situation.
Next, for the second paragraph, be more detailed about your career and awareness of problems.
If you dive deep into your knowledge of the current problems, it would be easier for you to answer the question " Why do you want to do that research in Japan or in your chosen university?"
Last but not the least, you should be able to write your reasons for wanting to be able to do your research in your chosen department. Your answer can revolve around the things you're able to do that not every laboratory or department can provide. What makes the laboratory different? You should be able to express that thought in the 3rd paragraph.
The details will vary from student to student, so we can only share the main points here.
